| Description: | The 5â² end of xTcof1 cDNA was amplified by PCR using BV1180 (5â²-ACTCAGATCCCATGGAGGAAGTGGAGGGATCC-3â²) and BV1181 (5â²-TCCAGAGCTCTCGAGTCACTGTGCAATCACAGGTGT-3â²) and subcloned into the NcoI and XhoI sites of pPROEX HTa vector. The E. coli-expressed poly-histidine-tagged protein, which included amino acids 1â307 of Xenopus treacle (xtreacle), was purified using Ni-NTA agarose (Qiagen, Inc.) and sent to Sigma for antibody production in chicken. |
